Start by checking the airflow. Is the vent in that room blowing about as strongly as the others? If it feels weak, there could be an issue with the ductwork or something blocking the airflow.

If the airflow feels normal, the room could be taking in more heat than the rest of the house. Large windows, poor insulation, or direct sunlight can all make a room harder to cool.
